    Experimental design: ANOVA? help!




    Hello,

    I have to analyze one paramater (lipid content) of seeds from 5 different cultivars. I am planning to do two independent analysis of the parameter from the 5 cultivar seeds, with 3 technical replicates for each independent analysis. I will have then 2 independent experiment with 3 technical replicates / each.

    1. Should I analyze the data using ANOVA?

    2. The "N" should be 2? (mean of the 3 replicates for the 2 independent experiments)
    or can I pool all the replicates from the 2 independent experiments and to have a N = 6?

    3 replicates for each of the 5 cultivars?
    And then do this another time?
    You can do it directly 6 replicates then.
    The questions of interest are other. The replicates are independent? What does technical replicate mean? The same subject is mearused 6 times? Give us more information

    From the grains of each cultivar I have to obtain the flours and then from those flours I extract the lipids and proteins. Then using a commercial kit I measure in triplicate (same day and time) the lipid and protein content that I have extracted. This whole process I name it as independent experiment 1.

    Then another day I do the same exact process (I take grains from the cultivars -> I obtain the flours -> I extract lipids and proteins ->I measure lipids and proteins content with commercial kit in triplicate in the same day and time). This is independent experimet 2.

    Then I have two independent measurements of lipids / proteins, with 3 "technical replicates" each.

    Is it ANOVA adequated for the analysis?

    The cultivars are the same each day or you take different ones?
    Where are the 3 replicates?

    It seems that mixed effects models are more suitable for this, but I am not sure yet what and how.
