PERMANOVA help with PRIMER-E

#1
Hi,

I am trying to look at differences in microbial communities at different elevations. I sampled at 4 sites, 2 at high elevation and 2 at low elevation. My sample sizes range 5 to 8 per site. In a MDS, the two high sites group together and the two low sites group together. If I run a PERMANOVA one for site and one for elevation as factors, separately, they are both highly significant. I tried to combine both factors (Site, Elevation, Site x Elevation) I get an error. Can you help me with the correct way to setup a PERMANOVA to determine if elevation is more important than site. I know this may be difficult since I do not have very much replication. I am using PRIMER-E to do this analysis.

Thanks!

Matt
